How to propagate Hypericum

Seeds are collected during the fruit ripening period from August to September each year. Well-growing Hypericum are selected for picking. After the capsule is dry, the seeds are rubbed out and the pure seeds are air-dried for later use. Generally, it is stored in bags at low temperature. Soaking the seeds in warm water for 1-2 days before sowing can significantly increase the germination rate of Hypericum seeds. It requires deep plowing and fine harrowing, applying sufficient base fertilizer, making a flat bed 1.0-1.5 meters wide, and waiting for sowing.

It can be sown in spring or autumn, usually in late March or early April. Choose windless weather, and it is best if there is no heavy rain or rainstorm 2-3 days after sowing. Either row sowing or broadcast sowing can be used. Cover the topsoil with about 1 cm, cover the bed with grass and sprinkle water on it. Seedlings will emerge 10 days after sowing and all will emerge in about 20 days.

Cuttings

It is usually carried out in spring or autumn. Choose strong 1-2 year old branches, cut them into 10-15 cm long cuttings with the cut ends flat on top and slanted on the bottom, and soak them in auxin or rooting powder. The Hypericum seedling bed should be watered thoroughly, and cuttings can be taken after the water has drained away. The spacing between rows and plants should be 10 cm × 10 cm, and the cutting depth should be 2/3 of the length of the cuttings.

Division

Division propagation is generally carried out during cultivation in spring or autumn. The division must be accompanied by a soil ball, which is conducive to survival. When digging up and dividing seedlings, you can wrap the Hypericum roots in nylon bags or plant them directly in pots. Try not to damage the root system of Hypericum. Dig and pack at the same time. After digging and packing, transport them in time and water them frequently to facilitate survival.

Tissue culture

Obtaining materials: Select young and tender branches of Hypericum sempervirens of the current year, rinse them in running water, cut them into 3 cm lengths, absorb excess water with filter paper, soak them in 70% alcohol for 20 seconds on a clean workbench, rinse them 3 times with sterile water, transfer them to 0.1% mercuric chloride solution and shake them for 5 minutes, rinse them 5 times with sterile water, then place them in a small magnetic disk covered with filter paper that has been sterilized by high pressure, cut them into small stem segments of 0.5-1.0 cm containing a bud, and inoculate them in differentiation medium for culture.

Cluster bud induction: After 20 days of incubation on differentiation medium, the sterile small stem segments gradually differentiate into cluster buds at the stem nodes, and after 30 days, the buds are 1-2.5 cm long. At this time, cut off the resulting clustered tender shoots, cut into small sections of about 1 cm and transfer to the propagation medium for culture. After 40 days, the average seedling height is 3-4 cm, and the proliferation coefficient is 6 to 8 times.

Rooting cultivation: When the Hypericum buds grow to 2-3 cm in height, cut off the tender stems and transfer them to rooting medium. After 2 weeks, each plant will have 3-5 roots, and the rooting rate reaches 99%. After the rooting, the test tube seedlings were placed in the greenhouse without opening the lid for 5 days, then opened for 2 days. They were taken out, washed, and soaked in clean water for 2-3 hours. They were transplanted into a matrix of vermiculite, river sand, and forest soil (5:1:3), covered with plastic film, and shade nets were set up to prevent direct sunlight. The humidity was maintained at around 90%. The plastic film was removed after 10 days, and new roots and leaves began to develop. The survival rate was 93.6%.
