Because under normal circumstances, its seeds are difficult to obtain, and artificial pollination cannot maintain the advantages of the mother plant. Therefore, tissue culture is an effective means. Specific stepsfirst stepChoose a Dendrobium officinale plant that is growing and developing normally, and take a relatively tender stem segment with five to six nodes. After taking it, rinse it under tap water for about an hour, remove all the leaves, and then cut it into two pieces. After cutting, soak it in 70 percent alcohol for about half a minute, and then sterilize it in raw mercury for six minutes. Next, rinse with sterile water several times, remove the leaf sheaths covering the stem segments, and then disinfect with raw mercury for three minutes. Then rinse with water and dry with paper. It is best to choose special absorbent paper. The water mentioned above must also be sterile. Finally, cut off the noded stem segments and stem tips, about one centimeter long. Step 2Place the above-mentioned stem segments and stem tips on the culture medium and wait for their side buds to begin to sprout (this can be observed in about eighteen days). When the apical and lateral buds grow to about one centimeter in length, cut off the young shoots and transfer them to another culture medium. After about three weeks, the buds will be about two centimeters long and will have two or three leaves. Step 3The first step in this step is to select the best developed and strongest young shoots for bud proliferation. Afterwards, if the length of the bud reaches about three centimeters, it can be induced. After cultivating for a period of time (about twenty days), about two to three roots will grow and a complete plant will be formed. Then let it continue to grow, and when it grows to two to three meters, the root system is almost very thick. Take it out of the culture medium and place it in the greenhouse, and it is considered a success. |
